2'-O-没食子酸金丝桃素(2'-O-GH)是从鹿蹄草中分离得到的一种活性化合物,具有显著的抗氧化活性。本研究的目的是研究2'-O-GH对氧化应激的肝脏保护作用,并阐明HepG2细胞和动物模型中潜在的机制信号通路。结果表明,2'-O-GH以剂量依赖性方式显著抑制过氧化氢(H2O2)诱导的HepG2细胞死亡。2'-OGH可显著抑制H2O2刺激的HepG2细胞中丝裂原活化蛋白激酶激活、活性氧生成、线粒体膜电位、细胞内钙水平以及随后的凋亡蛋白激活。这些结果提示2'-O-GH的肝保护作用可能与其抗氧化和自由基清除作用有关。

2'-O-galloylhyperin (2'-O-GH) is a bioactive compound isolated from Pyrola, with potent antioxidant activity. The aim of this study was to investigate the hepatoprotective effect of 2'-O-GH against oxidative stress, and to elucidate its underlying mechanistic signaling pathways in HepG2 cells and animal models. The results showed that 2'-O-GH significantly inhibited hydrogen peroxide (H₂O₂)-induced HepG2 cell death in a dose-dependent manner. 2'-O-GH also significantly inhibited mitogen-activated protein kinase (MAPK) activation, reactive oxygen species (ROS) generation, loss of mitochondrial membrane potential, intracellular calcium levels, and subsequent apoptotic protein activation in H₂O₂-stimulated HepG2 cells. These findings suggest that the hepatoprotective effect of 2'-O-GH may be associated with its antioxidant and free radical scavenging activities.
