This study comprehensively profiles the mSWI/SNF complexes throughout acute and chronic T cell stimulation, with and without small molecular inhibitors and degraders. Specifically, we find pan SWI/SNF and complex specific roles in both activation and exhaustion. Overall design: Comparative gene expression profiling analysis of RNA-seq data of CD3/CD28 activated Human Primary CD8 positive T cells with and without treatment of mSWI/SNF inhibitors or degraders and of CD3/CD28 activated Mouse Primary CD8 positive T cells with and without treatment with mSWI/SNF inhibitors.

本研究全面剖析了急性与慢性T细胞刺激过程中的mSWI/SNF复合物(mSWI/SNF complexes),并设置了小分子抑制剂、降解剂处理与未处理的对照组别。具体而言,本研究发现泛SWI/SNF复合物以及具有亚型特异性的复合物在T细胞活化与耗竭过程中均发挥关键调控作用。整体实验设计:对经CD3/CD28激活的人原代CD8阳性T细胞,分别施以mSWI/SNF抑制剂或降解剂处理与未处理;同时对经CD3/CD28激活的小鼠原代CD8阳性T细胞,分别施以mSWI/SNF抑制剂处理与未处理,对上述样本的RNA测序(RNA-seq)数据开展比较基因表达谱分析。
