To investigate the function of nuclear pore complex (NPC) in the regulation of zygotic genome activation (ZGA), we microinjected medium dosage of WGA in zebrafish embryos at 1-cell stage to block NPC function. We then performed gene expression profiling analysis using data obtained from RNA-seq of control or WGA treated embryo at comparable developmental time point (4.3 hours post fertilization (hpf) ) or develomental stage (dome). Comparative gene expression profiling analysis of RNA-seq data for zebrafish control embryo at 4.3 hpf of dome stage and WGA treated embryos at 4.3 hpf (developmental time matched) or 5 hpf (dome-like stage).
为探究核孔复合体(nuclear pore complex, NPC)在合子基因组激活(zygotic genome activation, ZGA)调控中的功能,我们于斑马鱼1细胞期胚胎中显微注射中等剂量的小麦胚芽凝集素(Wheat Germ Agglutinin, WGA)以阻断NPC的功能。随后,我们利用对照组与WGA处理组斑马鱼胚胎的RNA测序(RNA-seq)数据开展基因表达谱分析,两组胚胎的对应发育时间点为受精后4.3小时(hours post fertilization, hpf),对应发育阶段为圆顶期(dome)。本数据集针对斑马鱼对照组胚胎(4.3 hpf 或圆顶期)与WGA处理组胚胎的RNA-seq数据开展对比基因表达谱分析,其中处理组胚胎分别取自发育时间匹配的4.3 hpf样本,以及处于类圆顶期的5 hpf样本。
