为对比未成熟血小板与成熟血小板的转录组特征。方法:根据既往研究经验,将噻唑橙染色强度最高的15%血小板定义为网状血小板或未成熟血小板(reticulated or immature platelets, RPs),即RNA高表达组(RNAhigh);将噻唑橙染色强度最低的30%血小板定义为成熟血小板(mature platelets),即RNA低表达组(RNAlow),并对两组血小板进行分选(补充图S1D)。本研究共检测到1700余个差异表达基因,其中包含活化受体;未成熟血小板具有促血栓形成的转录组特征。实验总体设计:从健康供体中获取血小板RNA,并使用NextSeq-500平台进行测序分析。
