The histone methyltransferase Suv39h1 silences transcriptional programs during CD8+-T cell differentiation The study explores the role of gene silencing by Suv39h1, a histone methyltransferase, that plays a critical role in heterochromatin dynamics. Overall design: Naïve littermate and Suv39h1-KO CD8+ T cells, and day 7 p.i. Kb-OVA+ CD8+ T cells (from Listeria m.-OVA infected mice), were isolated by FACS. scRNA-seq was performed according to 10X Genomics technology. Both biological (Rep) and technical (tech) replicates were processed.
组蛋白甲基转移酶(histone methyltransferase)Suv39h1在CD8+T细胞分化过程中沉默转录程序。本研究探讨了组蛋白甲基转移酶Suv39h1介导的基因沉默作用,该酶在异染色质动态调控中发挥关键功能。实验整体设计如下:通过荧光激活细胞分选(FACS, Fluorescence-Activated Cell Sorting)分离未致敏同窝野生型CD8+T细胞、Suv39h1基因敲除(Suv39h1-KO)CD8+T细胞,以及经李斯特菌m.-OVA感染小鼠体内分离的感染后第7天Kb-OVA阳性CD8+T细胞。随后依照10X Genomics技术平台开展单细胞RNA测序(scRNA-seq, single-cell RNA sequencing)。实验同时设置了生物学重复(Rep)与技术重复(tech)并进行统一处理。
