肠致病性大肠杆菌 (ExPEC) 存在于肠道外时,会构成严重的人畜共患威胁,导致严重感染和各种宿主之间的潜在交叉传播。不断升级的抗菌素耐药性问题对临床细菌感染的治疗提出了挑战,需要制定创新策略。制定解决细菌感染的新方法至关重要。利用宿主作用抗菌化合物 (HAC) 的宿主导向疗法 (HDT) 已成为一种有价值的策略,通过关注宿主细胞内的宿主-病原体相互作用和细菌生存策略来应对细菌挑战。在这项研究中,我们进行了基于细胞的筛选,以确定能够抑制 ExPEC 在宿主细胞内存活和增殖的化合物。
Extraintestinal Pathogenic Escherichia coli (ExPEC) poses a severe zoonotic threat when residing outside the intestinal tract, causing serious infections and potential cross-transmission between various hosts. The escalating problem of antimicrobial resistance poses challenges to the treatment of clinical bacterial infections, calling for innovative strategies. It is critically important to develop novel approaches to combat bacterial infections. Host-directed therapy (HDT) that utilizes host-active antimicrobial compounds (HAC) has emerged as a valuable strategy to tackle bacterial challenges by focusing on host-pathogen interactions within host cells and bacterial survival strategies. In this study, we conducted a cell-based screen to identify compounds capable of inhibiting the survival and proliferation of ExPEC within host cells.
