L(3)mbt regulate the transcription of germ-specific genes in somatic cells but the detailed mechanism remains unclear. We performed ChIP-seq of L(3)mbt and a newly identified interactor in OSCs to elucidate the genome-wide localization. Then we performed RNA-seq of OSCs before and after RNAi treatment of L(3)mbt and its interactors and performed differential expression analysis. Overall design: ChIP-seq of OSCs and RNA-seq of OSCs before and after RNAi treatment of L(3)mbt and its interactors were performed in duplicate.

L(3)mbt 可调控体细胞内生殖特异性基因的转录,但其详细作用机制仍未阐明。我们针对卵巢体细胞干细胞(Ovary Somatic Cells, OSCs)中的L(3)mbt及其新鉴定的互作蛋白,开展了染色质免疫共沉淀测序(Chromatin Immunoprecipitation sequencing, ChIP-seq),以解析其全基因组定位特征。随后,我们对L(3)mbt及其互作蛋白实施RNA干扰(RNA interference, RNAi)处理,分别采集处理前后的OSCs进行转录组测序(RNA Sequencing, RNA-seq),并完成差异表达分析。整体实验设计:本研究针对OSCs开展了ChIP-seq实验,并对经L(3)mbt及其互作蛋白RNAi处理前后的OSCs进行了RNA-seq实验,所有实验均设置两次生物学重复。
