Microdissected ventromedial hypothalamus (VMH) was profiled to identify transcriptional changes after Nkx2-1 ablation in female mice. Total RNA was extracted from conditional mutants (Nkx2-1 f/f; Sf1Cre) and floxed controls (Nkx2-1 f/f) at postnatal day 10 (P10) and profiled using Mouse ref8 v2.0 Illumina chips and reagents (n=4/group). Overall design: The tissue profiled is microdissected ventromedial hypothalamus from P10 female mice. Controls and mutants are analyzed in quadruplicate.
本研究针对显微切割的腹内侧下丘脑(ventromedial hypothalamus, VMH)开展表达谱分析,以探究雌性小鼠Nkx2-1基因敲除后的转录组变化。实验于出生后第10天(postnatal day 10, P10),从条件性敲除突变体(Nkx2-1 f/f; Sf1Cre)及floxed对照(Nkx2-1 f/f)中提取总RNA,使用Mouse ref8 v2.0 Illumina芯片及配套试剂完成检测,每组设置4个生物学重复。
实验设计:本次分析的样本为出生后第10天雌性小鼠的显微切割腹内侧下丘脑,对照组与突变体组均设置4个生物学重复。
