The seminal proteome of a monandrous fly, Drosophila subobscura

We identify the seminal fluid proteins in a monandrous fly, Drosophila subobscura, using a label-free quantitative proteomics approach. The accessory glands and ejaculatory ducts of mated and unmated flies were dissected. Tissues from 10 males were pooled for each sample of mated and unmated males and there were 5 samples per mating treatment. We identified proteins that had significantly higher abundance in unmated than mated males. These proteins were likely to be transferred to females during mating and so represent the candidate seminal fluid proteins.

本研究采用无标记定量蛋白质组学（label-free quantitative proteomics）方法，对单雌交配型暗果蝇（Drosophila subobscura）的精浆蛋白（seminal fluid proteins）进行鉴定。本研究解剖了交配组与未交配组果蝇的附腺和射精管；交配组与未交配组的每个样本均混合10只雄蝇的组织，且每个交配处理设置5个重复样本。本研究鉴定出未交配雄蝇中丰度显著高于交配雄蝇的蛋白质，这类蛋白质极有可能在交配过程中转移至雌蝇体内，因此可作为候选精浆蛋白。
