This project presents the haplotype-resolved reference genome of Eidothea hardeniana (Nightcap Oak), a critically endangered tree species in the family Proteaceae. Endemic to the Nightcap Range of north-eastern New South Wales, Australia, E. hardeniana is a paleo-endemic rainforest tree of high conservation and evolutionary significance. The genome was assembled using PacBio HiFi, Oxford Nanopore, and Hi-C sequencing, providing chromosome-level haplotypes. These resources will support studies of genome evolution, genetic diversity, and conservation management in the Proteaceae and other ancient angiosperm lineages.

本研究报道了宽柄叶木(*Eidothea hardeniana*,俗称夜帽橡树(Nightcap Oak))的单倍型解析参考基因组(haplotype-resolved reference genome)。该物种为山龙眼科(Proteaceae)极度濒危的乔木类群,仅分布于澳大利亚新南威尔士州东北部的夜帽岭(Nightcap Range),是兼具极高保护价值与进化研究意义的古特有雨林乔木。本研究采用PacBio HiFi、牛津纳米孔(Oxford Nanopore)及Hi-C测序技术完成基因组组装,获得了染色体级别的单倍型序列,上述基因组资源将为山龙眼科及其他古老被子植物演化支的基因组进化、遗传多样性与保护管理研究提供关键支撑。
