Popillia japonica (Coleoptera: Scarabaeidae) is an invasive pest of Europe and America originally from Japan. In the Azores, this pest was first found on Terceira Island during the sixties and spread to other regions of the nine islands of the Archipelago where it becomes established mostly in pastures. The rate of infestation differs between sites despite the type of vegetation and weather conditions being similar. Aiming to find drivers explaining these differences we hypothesize microbiome could play a role. Therefore, we sampled larvae and soil from sites with high and low rates of infestation to analyze for microbiome by Next-Generation Sequencing. We sequenced twenty four 16S DNA libraries producing 2.445,131 raw input reads which resulted in 598,595 valid reads after checking and denoising raw reads.
日本丽金龟(Popillia japonica)隶属于鞘翅目(Coleoptera)金龟科(Scarabaeidae),是原产于日本、现已入侵欧美地区的外来入侵害虫。在亚速尔群岛,该害虫于20世纪60年代首次在特塞拉岛被发现,随后扩散至该群岛九个岛屿的其余区域,且主要在牧场中定殖。尽管各采样点的植被类型与气候条件相近,但不同采样点的虫害发生率仍存在显著差异。为阐明造成这种虫害发生率差异的驱动因子,本研究提出微生物组可能参与其中的假说。因此,本研究从虫害发生率高低不同的两类采样点采集幼虫与土壤样本,采用下一代测序(Next-Generation Sequencing)技术分析其微生物组。本研究共构建24个16S DNA文库,初始原始测序reads总数达2,445,131条,经原始数据质控与去噪处理后,最终获得有效reads 598,595条。
