Origin of the public memory antibody response induced upon antiviral vaccination in fish

In this work, we studied the modifications of the Ab repertoires following vaccination with an attenuated VHSV in rainbow trout. To this purpose, we set up a cDNA barcoded strategy to sequence the repertoire of IgM and IgT heavy chain transcripts corresponding to selected VH families. We show that the persisting memory response contains public IgM clonotypes. The vaccination also induces private responses for both IgM and the mucosal IgT, implicating clonotypes that are rare in naïve fish. This study provides an evolutionary perspective on the development of public antibody specificities potentially ensuring universal protection after infection.
本研究针对虹鳟接种减毒病毒性出血性败血症病毒(VHSV)后抗体库的动态变化展开探究。为此,我们构建了一套cDNA条形码测序策略,对选定的VH家族对应的IgM与IgT重链转录本的抗体库进行测序分析。研究结果显示,持续存在的记忆性免疫应答中包含公共型IgM克隆型。该疫苗接种同时可诱导IgM与黏膜型IgT产生私有型免疫应答,涉及在未致敏鱼类体内较为稀少的克隆型。本研究为公共型抗体特异性的演化发育提供了全新视角,这类特异性有望在感染后提供广谱保护性免疫。
