Five critical life-endangered COVID-19 patients were enrolled to donate blood. Family signed consent was obtained. The peripheral blood leukocytes were obtained by standard centrifugation methods. Then the leukocytes were single cell RNA sequenced using 10X Genomics Protocol. High presence of myeloid lineage subsets whereas low count of lymphoid lineage populations in COVID-19 patients was readily observed. Interestingly, among the cellular subset present in myeloid lineage there was immature states.
招募5名危重症新型冠状病毒肺炎(COVID-19)患者参与献血,已获取其家属签署的知情同意书。采用标准离心法分离获取外周血白细胞,随后基于10X Genomics技术流程对白细胞开展单细胞RNA测序。可直观观察到,COVID-19患者体内髓系谱系亚群占比显著偏高,而淋巴细胞谱系细胞群数量偏低。值得注意的是,在髓系谱系的细胞亚群中,还存在未成熟状态的细胞群体。
