Focusing on the Lake Kasumigaura watershed, we aimed to detect and assess trade-offs and synergies between multiple ecosystem services and biodiversity in order to receive ecosystem services sustainably. The Lake Kasumigaura watershed was divided into 50 sub-watersheds, and ecosystem services such as water quality and land use were examined in each watershed, as well as fish diversity as an indicator of biodiversity. Water quality and fish diversity were surveyed twice a year, in summer and winter, from 2016 to 2019. In this project, we investigated the fish diversity in each watershed regarding the use of environmental DNA extracted from river water. Metabarcoding was performed on amplicons using universal primers that amplify fish mitochondrial 12S rRNA (Miya et al. 2015).
本研究以霞浦湖(Lake Kasumigaura)流域为研究对象,旨在探测并评估多重生态系统服务(ecosystem services)与生物多样性(biodiversity)之间的权衡关系与协同效应,以实现生态系统服务的可持续利用。本研究将霞浦湖流域划分为50个子流域,针对每个子流域开展了水质(water quality)、土地利用(land use)等生态系统服务的调查,并以鱼类多样性作为生物多样性的指示指标。2016年至2019年间,研究团队于每年夏季与冬季分两次开展水质与鱼类多样性的野外调查。本项目依托从河水中提取的环境DNA(environmental DNA, eDNA),对各子流域的鱼类多样性展开了调查。本研究采用可扩增鱼类线粒体12S核糖体RNA(mitochondrial 12S rRNA)的通用引物(universal primers),对扩增子(amplicon)进行了元条形码测序(metabarcoding),相关方法参见Miya等(2015)的研究。
