Identification of genes associated with bendiocarb resistance. Mosquitoes collected as larvae from Nagongera and Kihihi, Uganda. Bendiocarb-resistant and unexposed female mosquitoes selected using standard WHO tube bioassays. RNA was extracted from pools of five individuals identified as An. gambiae s.s. Insecticide-susceptible mosquitoes from the Kisumu strain were included as controls. RNA hybridized in an interwoven loop design to compare four biological replicates each of resistant, unexposed, and laboratory mosquitoes.
本研究旨在鉴定与苯威(bendiocarb)抗性相关的基因。实验所用蚊虫采自乌干达纳贡杰拉(Nagongera)与基希希(Kihihi)的幼虫种群。采用世界卫生组织(WHO)标准筒式生物测定法,筛选出苯威抗性与未接触药剂的雌性蚊虫。从经鉴定为冈比亚按蚊(An. gambiae s.s.)的5只个体混合样本中提取RNA。以基苏姆(Kisumu)品系的杀虫剂敏感型蚊虫作为对照。采用交错循环设计对RNA进行杂交,分别比较抗性组、未接触组及实验室饲养组蚊虫的4次生物学重复样本。
