In order to explore the effect of betaine on NAFLD and its mechanism, we divided mice into high-fat diet (HF) and high-fat diet with betaine (HB) groups, and used N6-methyladenosine sequence (m6A-seq), RNA-seq and mRNAs bound to ribosome-nascent chain complex sequencing (RNC-seq) to evaluate the gene expression levels in the liver of each group. Examination of anti-m6A antibody in liver tissue. Total RNA and ribosome-bound RNA were extracted from the livers of high-fat diet- (HF) and High-fat diet plus Betaine (HB)-fed mice, and the polyA+ mRNA was sequenced using Illumina HiSeq X Ten sequencer. The reads were mapped to RefSeq RNA reference sequences and the expression level were quantified by rpkM calculation.
为探究甜菜碱对非酒精性脂肪性肝病(Non-Alcoholic Fatty Liver Disease,NAFLD)的作用及其潜在机制,本研究将小鼠分为高脂饮食(HF)组与高脂饮食添加甜菜碱(HB)组,并采用N6-甲基腺苷测序(m6A-seq)、RNA测序(RNA-seq)以及核糖体新生肽链复合物结合mRNA测序(RNC-seq)技术,对各组小鼠肝脏的基因表达水平进行评估,同时对肝组织中的抗m6A抗体开展检测。本研究从高脂饮食(HF)组与高脂饮食添加甜菜碱(HB)组饲喂的小鼠肝脏中提取总RNA及核糖体结合RNA,并利用Illumina HiSeq X Ten测序仪对其中的polyA+ mRNA进行测序;将测序读段比对至RefSeq RNA参考序列,通过RPKM计算对基因表达水平进行定量。
