Vibrio cholerae containing the phage inducible chromosomal island-like element (PLE) were infected with the lytic Vibrio cholerae phage ICP1. ICP1 infection triggers transcriptional activation of PLE, which we leverage here to study the RNA-RNA interactome of ICP1 and PLE. Hi-GRIL-seq was carried out in the system, leading to in vivo ligation of interacting transcripts which were subsequently isolated and sequenced with paired end 150x150 RNAseq, allowing for identification of interacting transcripts during ICP1 infection.
携带噬菌体诱导性染色体岛样元件(PLE)的霍乱弧菌被裂解性霍乱弧菌噬菌体ICP1感染。ICP1感染可触发PLE的转录激活,本研究借此探究ICP1与PLE的RNA-RNA互作组。在该实验体系中开展了Hi-GRIL-seq实验,实现了互作转录本的体内连接,随后对其进行分离并采用150×150双端RNA测序,从而能够鉴定ICP1感染过程中的互作转录本。
