Here we determine the genome-wide binding sites of PRMT1 in primary human keratinocytes. We also investigated the impact of casein kinase inhibitor D4476 on PRMT1 genomic binding. Overall design: HA-PRMT1 was expressed at near endogenous level in primary keratinocytes. Two independent HA antibodies were used for PRMT1 ChIP-seq. The CSNK1a1 inhibitor D4476 was added to keratinocyte culture to characterize the role of CSNK1a1 on controlling PRMT1 genomic binding.
本研究鉴定了原代人角质形成细胞中蛋白质精氨酸甲基转移酶1(Protein Arginine Methyltransferase 1, PRMT1)的全基因组结合位点,同时探究了酪蛋白激酶抑制剂D4476对PRMT1基因组结合的影响。整体实验设计如下:将HA-PRMT1(HA标签,Hemagglutinin tag)以接近内源水平的方式在原代角质形成细胞中表达;采用两种独立的HA抗体开展PRMT1的染色质免疫共沉淀测序(Chromatin Immunoprecipitation sequencing, ChIP-seq)实验;向角质形成细胞培养液中添加酪蛋白激酶1α1(Casein Kinase 1 alpha 1, CSNK1a1)抑制剂D4476,以阐明CSNK1a1在调控PRMT1基因组结合过程中的作用。
