Comparison of transcript abundance between control (untreated) and methotrexate treated S3 Drosophila cells and ovaries disected from female flies. Keywords: Stress response Overall design: S3 cells were exposed to 0 or 5.2 X 10^-8 M MTX for 4 days then harvested. Flies were exposed to 0 or 5 ppm MTX for 5 days then ovaries were dissected. RNA was extracted from S3 cells and ovaries. 4 S3 microarrays with one dye-swap and 2 ovary microarrays swapping dyes were hybridized to spotted cDNA microarrays.
本研究对对照组(未处理组)与甲氨蝶呤(methotrexate, MTX)处理的S3果蝇细胞,以及雌性果蝇解剖获取的卵巢的转录本丰度进行了比较。

实验设计:将S3细胞分别置于不含或含5.2×10^-8 M甲氨蝶呤的培养基中培养4天后收集样本;将雌性果蝇分别暴露于不含或含5 ppm甲氨蝶呤的环境中5天后,解剖分离卵巢。分别从S3细胞和卵巢中提取RNA,采用斑点cDNA微阵列(cDNA microarray)进行杂交实验:其中S3细胞组设置4张微阵列,包含1次染料互换;卵巢组设置2张微阵列,均采用染料互换方案。
