To analyze the contribution of dietary PUFAs and adipocyte LPCAT3 expression in modulating the transcriptome of murine inguinal white adipose tissue during adipocyte hypertrophy. 本研究旨在解析膳食多不饱和脂肪酸(Polyunsaturated Fatty Acids, PUFAs)与脂肪细胞溶血卵磷脂酰基转移酶3(Lysophosphatidylcholine Acyltransferase 3, LPCAT3)的表达,在脂肪细胞肥大过程中对小鼠腹股沟白色脂肪组织(inguinal white adipose tissue, iWAT)转录组的调控作用。本研究采用双因素实验设计开展批量RNA测序。实验分为野生型(Wild Type, WT)与AdL3基因敲除(AdL3KO)两种小鼠品系,分别饲喂脂肪供能占比60%的富含ω-6的高脂饲料,干预时长分别为0周(断奶后饲喂常规饲料)、1周、5周或10周。实验设计概述:对饲喂上述脂肪供能占比60%的富含ω-6高脂饲料、干预时长分别为0周(断奶后饲喂常规饲料)、1周、5周或10周的WT与AdL3KO小鼠的iWAT组织进行比较基因表达谱分析。
