Gene alteration in A549 cells by disinfectant PHMG

Polyhexamethyl guanidine is widely used as a disinfectant in various places such as hospitals and restaurants. In order to examine of PHMG cytotoxicity in lung adenocarcinoma A549 cells we treated 5 ug/ml PHMG for 24 h. To investigate cytotoxicity of commonly used bactericidal reagent PHMG, A549 cells were treated 5 ug/ml PHMG for 24 h. After then purified the total RNA from treated and untreated cells using the Rneasy mini kit (Qiagen). The concentration and purity of the total RNA was measured and performed gene array.

聚六亚甲基胍（Polyhexamethyl guanidine，PHMG）作为消毒剂被广泛应用于医院、餐厅等各类场所。为探究PHMG对肺腺癌A549细胞的细胞毒性，我们采用5 μg/ml的PHMG处理细胞24小时。为进一步考察常用杀菌试剂PHMG的细胞毒性，我们以5 μg/ml的PHMG处理A549细胞24小时。随后，我们使用RNeasy Mini试剂盒（Qiagen）从经PHMG处理的细胞及未处理的对照细胞中提取总RNA。我们测定了总RNA的浓度与纯度，并开展了基因芯片（gene array）分析。
