A DNA library with glycyl–histidine-functionalized tertiary amine moiety modification at the cleavage junction was used for in vitro selection of Zn2+-dependent DNAzyme. Four DNAzymes were isolated and each of these DNAzymes displays different cooperative binding of metal ions. An interesting relationship between the number of metal ions involved in DNAzyme catalysis and selectivity was established, and this study showed that it could be a useful way to develop highly specific metal sensors.
本研究采用在切割位点修饰有甘氨酰组氨酸功能化叔胺基团的DNA文库,用于锌离子(Zn²+)依赖型脱氧核酶(DNAzyme)的体外筛选。最终筛选获得4种脱氧核酶,每一种均展现出差异化的金属离子协同结合行为。本研究确立了脱氧核酶催化过程中所涉及的金属离子数量与其选择性之间的有趣关联,证实该策略可作为开发高特异性金属传感器的有效路径。
