This study aimed to identify oxidoreductase genes involved in biosynthesis of 20-hydroxyecdysone(20E) in spinach (Spinacia oleracea), a plant that contains high levels of 20E. To achieve this, spinach samples with varying 20E contents were generated: 20E-positive wild-type spinach, 20E-negative callus derived from spinach roots, spinach with an increased 20E content due to pds gene knockdown via the virus-induced gene silencing system, and its control, and then RNA sequencing (RNA-seq)-based comparative transcriptome analysis was performed.
本研究旨在鉴定菠菜(Spinacia oleracea)中参与20-羟基蜕皮甾酮(20-hydroxyecdysone,20E)生物合成的氧化还原酶基因;菠菜是一种20E含量较高的植物。为达成研究目标,我们制备了20E含量各异的菠菜样本:20E阳性野生型菠菜、源自菠菜根系的20E阴性愈伤组织、通过病毒诱导基因沉默系统敲低pds基因以提升20E含量的菠菜及其对照样本,随后开展了基于RNA测序(RNA sequencing,RNA-seq)的比较转录组分析。
