We proposed to uncover the RNome of a saprophytic strain ofL. biflexa, by a systematic RNomic approach. Total transcriptome of non-pathogenic L. biflexa will be identified through differential(dRNA-seq) transcriptomic mapping. TSS profile and differential RNA expression will be annotated. Importantly, we will compare thetranscriptome profile of L. biflexa with those of L. interrogans to identify the RNA candidates (mRNA and regulatory sRNA) that areunique to pathogenic and saprophytic leptospira.
本研究拟通过系统性RNA组学方法,解析双曲钩端螺旋体(Leptospira biflexa, L. biflexa)腐生菌株的RNA组(RNome)。将通过差异RNA测序(differential RNA-seq, dRNA-seq)转录组图谱分析,鉴定非致病双曲钩端螺旋体的完整转录组,并注释其转录起始位点(Transcription Start Site, TSS)谱与差异RNA表达情况。尤为关键的是,本研究将对比双曲钩端螺旋体与问号钩端螺旋体(Leptospira interrogans, L. interrogans)的转录组特征,以筛选出致病型与腐生型钩端螺旋体各自特有的RNA候选分子(信使RNA (messenger RNA, mRNA) 与调控型小RNA (small RNA, sRNA))。
