Cannabidiol (CBD) is a phytocannabinoid known for its anti-inflammatory, immunomodulatory, antioxidant, anxiolytic, antipsychotic, and anticonvulsant effects. The systemic administration of CBD also has the potential to impact kidney function, either beneficially or adversely. This study aims to examine the effects of CBD on behavior, immune system activity, and stress levels, and to determine its impact on transcriptomic alterations in selected organs of mice. (Study funded by the National Science Centre, Poland; project no. 2020/39/O/NZ9/00821) Overall design: Male mice of the standard C57BL/6J strain were used. The animals were added to the long-term intraperitoneal CBD administration (28 days). There were two subgroups receiving different doses of CBD - medium (10 mg/kg bw) and high, used in some clinical trials (20 mg/kg bw). Additionally, a control group (placebo/vehicle) was maintained. Chemically pure Cannabidiol (CBD) was administered intraperitoneally (in the research group) to ensure that the substances reached the blood vessels of the digestive system. CBD was administered in a single daily dose. The mice were housed under standard conditions and with standard feeding and watering ad libitum. They were also examined every third day to determine well-being and health. After the period of CBD administration, the mice were immediately euthanized and organs such as the brain, spleen and liver, kidneys, and others were collected. In this part of the experiment, kidney samples were used to isolate the total RNA. The obtained RNA was quality assessed and used to prepare directional libraries for high throughput transcriptome sequencing using CORALL mRNA-Seq V2 kit (Lexogen). The libraries were sequenced commercially in a 2x150bp paired-end run. The obtained sequence data were used to analyze the differential expression between the different CBD dosages and the control group. Differentially expressed genes were classified into particular processes or functions and were subjected to statistical testing for overrepresentation. The synthesis of the obtained data will allow to propose of potential mechanisms of CBD action and create an atlas of changes taking place in the transcriptome of the main target organs. 实验整体设计:本实验选用标准品系C57BL/6J雄性小鼠,所有小鼠均接受为期28天的长期腹腔注射给药处理,其中分为两个CBD给药亚组,分别给予中等剂量(10 mg/kg体重)与部分临床试验中采用的高剂量(20 mg/kg体重);另设置安慰剂/溶媒对照组。为确保药物经消化系统血管吸收入血,本实验采用化学纯级别的大麻二酚对给药组小鼠进行腹腔注射,给药方式为每日单次注射。小鼠饲养于标准实验环境中,自由采食与饮水,且每三日进行一次健康状况与状态评估。给药周期结束后,立即对小鼠实施安乐死,并采集脑、脾脏、肝脏、肾脏等多个器官样本。本实验部分将采用肾脏样本提取总RNA,对提取得到的RNA进行质量检测后,使用Lexogen公司的CORALL mRNA-Seq V2试剂盒构建定向转录组测序文库,随后以2×150bp双端测序模式开展商业化高通量转录组测序。所得序列数据将用于分析不同CBD剂量组与对照组间的差异表达基因。将差异表达基因归类至特定生物学过程或功能通路,并进行富集统计学检验。本研究通过整合分析所得数据,有望阐明大麻二酚发挥作用的潜在分子机制,并构建主要靶器官转录组变化图谱。(本研究由波兰国家科学中心资助,项目编号:2020/39/O/NZ9/00821)
