Accuracy of allele frequency estimation from pooled RNA-seq. Myodes glareolus

Bank vole liver transcriptomes were used for testing accuracy of allele frequency estimation from pooled RNA-seq sample. Each sample was sequenced both as an indivudually barcoded library and as a part of a pool. General aim was to determine how various aspects of expression level variation influence allele frequency estimation.

本研究以银行田鼠（Bank vole）肝脏转录组为研究材料，用于验证基于混合RNA测序（pooled RNA-seq）样本的等位基因频率估计（allele frequency estimation）精度。每份样本均采用两种测序方式：一是构建单独条形码标记文库（individually barcoded library）进行测序，二是以混合样本池的组成部分完成测序。本研究的总体目标为明确表达水平变异的各维度如何对等位基因频率估计产生影响。
