The WDR-23 protein regulates the transcription factor SKN-1 directly. C. elegans wdr-23 mutants have highly active SKN-1 and are stress resistant, long-lived, small, and reproduce poorly. We used microarrays to measure global gene expression in wdr-23 mutants and to indentify genes regulated by SKN-1. RNA extraction and hybridization on Affymetrix microarrays was performed on three genetic backgrounds at the L4 larval stage. The backgrounds were N2 (wildtype) with control RNAi, wdr-23(tm1817) mutants with skn-1 RNAi, and wdr-23(tm1817) mutants with control RNAi.
WDR-23蛋白(WDR-23 protein)可直接调控转录因子SKN-1(SKN-1)。秀丽隐杆线虫(Caenorhabditis elegans)的wdr-23基因突变体中SKN-1活性极高,且具备抗逆、长寿、体型偏小及繁殖力低下的表型。本研究采用基因芯片(microarrays)检测wdr-23基因突变体的全基因组表达谱,以鉴定受SKN-1调控的基因。实验于L4幼虫期,对三种遗传背景的样本开展RNA提取及Affymetrix基因芯片杂交实验,三种遗传背景分别为:携带对照RNA干扰(RNA interference, RNAi)的野生型N2菌株、经skn-1 RNAi处理的wdr-23(tm1817)基因突变体,以及携带对照RNAi的wdr-23(tm1817)基因突变体。
