Intravenous administration of stable isotope labeled amino acid ((13)C6-leucine) to humans recently made it possible to study the metabolism of specific biomarkers in cerebrospinal fluid (CSF) and in plasma using targeted mass spectrometry (MS). This labeling approach could be of great interest for monitoring many leucine-containing peptides in parallel, using Mass Spectrometry. This will make it possible to quantify the rates of synthesis and clearance of a large range of proteins in humans with a view to obtaining new insights into protein metabolism processes and the pathophysiology of diseases such as Alzheimer's disease.
向人体静脉输注稳定同位素标记的氨基酸((13)C6-亮氨酸),近期已使得借助靶向质谱(targeted mass spectrometry,MS)技术研究脑脊液(cerebrospinal fluid,CSF)与血浆中特定生物标志物的代谢情况成为可能。此种同位素标记策略可通过质谱技术同步监测多种含亮氨酸的肽段,具备极高的应用潜力;此举将可量化人体中大量蛋白质的合成与清除速率,以期为解析蛋白质代谢过程以及阿尔茨海默病(Alzheimer's disease)等疾病的病理生理机制提供全新见解。
