Aac11/Api5 is a conserved anti-apoptotic protein that functions through a poorly understood mechanism that appears to involve splicing and transcription. Here we used a HTS approach to identify RNAs that respond to Aac11/Api5 loss in whole 3rd instar larvae. Overall design: An Aac11/Api5 UAS-RNAi transgene was used to deplete the mRNA and its encoded protein from third instar larvae using the Actin5c-Gal4 driver.

Aac11/Api5是一种保守的抗凋亡蛋白,其功能发挥依赖于一套尚未阐明的分子机制,现有研究提示该机制可能涉及剪接与转录过程。本研究采用高通量筛选(High-Throughput Screening, HTS)策略,在完整三龄幼虫中鉴定出响应Aac11/Api5基因缺失的RNA分子。 总体实验设计:通过Actin5c-Gal4驱动因子,利用Aac11/Api5的UAS-RNAi转基因元件对三龄幼虫的mRNA及其编码蛋白进行敲低。
