Identification of the mitochondrion-targeted ZmPPR67 substrate RNA using Hiten strategy

By using Hiten method, mitochondrial atp9 was identified as the substrate RNA of ZmPPRR67. This was verified by multiple downstream experiments including cRT-PCR, 5'RACE, RNA gel blot, BN PAGE, and Western blot.

本研究采用Hiten法，鉴定出线粒体atp9为ZmPPRR67的底物RNA。上述结论经多项下游实验验证，涵盖竞争性逆转录聚合酶链式反应（competitive Reverse Transcription PCR，cRT-PCR）、5'末端快速扩增（5' Rapid Amplification of cDNA Ends，5'RACE）、RNA凝胶印迹杂交（RNA gel blot）、蓝色天然聚丙烯酰胺凝胶电泳（Blue Native Polyacrylamide Gel Electrophoresis，BN PAGE）及蛋白质印迹（Western blot）。
