Gene expression signature of EpCAMlow/PDPN+ and EpCAMhigh/PDPN- cancer cells in engineered human NSCLC HSAEC_4T53RD mouse xenograft model. Overall design: HSAEC_4T53RD cells were subcutaneously inoculated into BALB/cSlc-nu/nu mice. Tumor cells were digested and mouse leukocyte and stromal cells were eliminated by using microbieads. Cancer cells were stained by using anti-EpCAM antibody and anti-PDPN antibody, and EpCAMlow/PDPN+ and EpCAMhigh/PDPN- cells were collected by FACS. Gene expression analysis was conducted using Agilent microarray.

本数据集为工程化人类非小细胞肺癌(NSCLC)HSAEC_4T53RD小鼠异种移植模型中EpCAMlow/PDPN+与EpCAMhigh/PDPN-癌细胞的基因表达特征。整体实验设计:将HSAEC_4T53RD细胞皮下接种至BALB/cSlc-nu/nu免疫缺陷小鼠体内;对肿瘤组织进行消化,通过免疫磁珠去除小鼠白细胞及基质细胞;使用抗上皮细胞黏附分子(EpCAM)抗体与抗Podoplanin(PDPN)抗体对残留的肿瘤细胞进行染色,借助荧光激活细胞分选术(Fluorescence-Activated Cell Sorting, FACS)分选出EpCAMlow/PDPN+与EpCAMhigh/PDPN-癌细胞;最终采用安捷伦(Agilent)基因芯片开展基因表达分析。
