We used genetic approaches to identify butyrophilin 3A1 (BTN3A1) as the molecule that binds and presents phosphorylated antigens to Vγ9Vδ2 TCR–expressing cells. BTN3A1 represents a novel antigen–presenting molecule for the activation of Vγ9Vδ2 TCR–expressing cells which play important roles in human tumor immunity and host defense. Overall design: 3 replicates of 4 cell lines (12 samples total) were analysed. Data was RMA normalized, and genes that expressed above the negative control in all 4 cell lines from chromosome 6 considered for further analysis
本研究通过遗传学方法鉴定出嗜乳脂蛋白3A1(BTN3A1),该分子可结合并呈递磷酸化抗原至表达Vγ9Vδ2 T细胞受体(T cell receptor, TCR)的细胞。嗜乳脂蛋白3A1是一类新型抗原呈递分子,能够激活表达Vγ9Vδ2 TCR的细胞;此类细胞在人类肿瘤免疫与宿主防御过程中发挥关键作用。
实验设计:本研究对4种细胞系的3次生物学重复样本(总计12个样本)进行了分析。
数据经RMA(Robust Multi-array Average,稳健多数组平均)归一化处理后,本研究对来自6号染色体、且在全部4种细胞系中表达量均高于阴性对照的基因开展了后续分析。
