Liver has a crucial role in the regulation of immune defense in systemic infections. During endotoxemia, the liver transits from an immune-tolerant towards an immune-active state and forms the first line of defense against invading microorganisms. the role of liver nonparenchymal cells in endotoxemia remains unintelligible. To characterize the liver nonparenchymal cells in endotoxemia liver, we performed single-cell RNA sequencing of liver nonparenchymal cells from healthy C57BL/6J mice and murine model of endotoxemia at early or late stage. The single-cell RNA-seq analyses revealed the heterogeneity of liver nonparenchymal cells in endotoxemia liver. Overall design: Single-cell RNA sequencing of liver nonparenchymal cells from healthy C57BL/6J mice (n=6, eight weeks-age) using MGISEQ-2000. Single-cell RNA sequencing of liver parenchymal cells from murine model of endotoxemia at 3-hour time point (n=6, eight weeks-age) using MGISEQ-2000. Single-cell RNA sequencing of liver parenchymal cells from murine model of endotoxemia at 12-hour time point (n=6, eight weeks-age) using MGISEQ-2000.
肝脏在全身感染的免疫防御调控中发挥关键作用。在内毒素血症(endotoxemia)期间,肝脏会从免疫耐受状态转向免疫激活状态,构成抵御入侵微生物的第一道防线。目前肝脏非实质细胞(liver nonparenchymal cells)在内毒素血症中的作用仍尚未阐明。为解析内毒素血症肝脏中的肝脏非实质细胞特征,我们对健康C57BL/6J小鼠以及早、晚期内毒素血症小鼠模型的肝脏非实质细胞开展了单细胞RNA测序(single-cell RNA sequencing)。单细胞RNA测序分析揭示了内毒素血症肝脏中肝脏非实质细胞的异质性。整体实验设计:使用MGISEQ-2000平台对健康C57BL/6J小鼠(n=6,8周龄)的肝脏非实质细胞进行单细胞RNA测序;使用MGISEQ-2000平台对3小时时间点的内毒素血症小鼠模型(n=6,8周龄)的肝脏实质细胞(liver parenchymal cells)进行单细胞RNA测序;使用MGISEQ-2000平台对12小时时间点的内毒素血症小鼠模型(n=6,8周龄)的肝脏实质细胞进行单细胞RNA测序。
