本研究采用RNA测序(RNA-seq)技术,鉴定sir2Δhst1Δ菌株与野生型(wild-type)菌株间的差异表达基因。本次测序的菌株为携带hmr基因缺失的matα菌株,以避免假二倍体(pseudodiploid)状态。本研究选取sir2Δhst1Δ双缺失菌株进行差异表达分析,以规避单基因缺失菌株中可能存在的基因互补效应。本实验为每种菌株设置了两次技术重复(technical replicates)。
