This study was to identify the role of miR-433 and to clarify the underlying molecular mechanisms in cardiac remodeling post myocardial infarction. Overall design: To determine if inhibition of miR-433 is sufficient to prevent cardiac fibrosis in vivo, mice were injected via tail vein with 80 mg/kg miR-433 antagomir or the scramble control for 3 consecutive days and subjected to left anterior descending coronary artery (LAD) ligation.Twenty-one days post myocardial infarction, gene arrays were conducted to compare the difference of gene expressions between miR-433 antagomir (n=3) or scrambled negative control (n=3) post myocardial infarction.
本研究旨在明确miR-433的作用,并阐明心肌梗死(myocardial infarction)后心脏重构的潜在分子机制。
实验设计:为验证miR-433抑制是否足以在体内预防心脏纤维化,本研究通过尾静脉向小鼠注射80 mg/kg的miR-433拮抗剂(antagomir)或错配阴性对照序列,连续给药3天后,对小鼠实施左前降支冠状动脉(left anterior descending coronary artery,LAD)结扎术。心肌梗死后21天,通过基因芯片(gene arrays)检测比较miR-433拮抗剂组(n=3)与错配阴性对照组(n=3)的基因表达差异。
