TMEM41B is a host factor required for the replication of diverse coronaviruses including SARS-CoV-2

Identification of a host factor essential for coronavirus replication A CRISPR/Cas9 library was used to identify genes that rendered the cell refractory to killing by hCoV-229E. The complete sgRNA librarywas packaged into lentivirus and used to transduce Huh7 cells . In two independent biological replicates, the transduced cells were infected with the virus, and the cells that were not killed by infection were collected and the sgRNAs enriched in those populations were sequenced.

冠状病毒复制必需宿主因子的鉴定。本研究利用CRISPR/Cas9文库（CRISPR/Cas9 library）筛选可使细胞对hCoV-229E（人冠状病毒229E）杀伤产生抵抗的基因。将完整的sgRNA（单引导RNA）文库包装至慢病毒（lentivirus）中，用于转导Huh7细胞。在两次独立的生物学重复实验中，将转导后的细胞进行病毒感染，收集未被病毒杀伤的细胞，并对该细胞群中富集的sgRNA进行测序。
