In the present study, RNA sequencing was performed to explore the lncRNA and mRNA expression profiles in lung tissue of WT PCP mice and BAFF-R PCP mice. Additionally, to identify the underlying biological functions of differentially expressed RNAs, bioinformation analysis including gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis and lncRNA-mRNA co-expression network construction was conducted. We furtherly explored the role of mature B cells in the pathogenesis and progression of PCP and the data demonstrated that BAFF-R deficiency play a significant role in immune regulation in PCP population.

本研究通过RNA测序(RNA sequencing),探究野生型(Wild Type, WT)肺孢子菌肺炎(Pneumocystis pneumonia, PCP)小鼠与BAFF-R(B细胞活化因子受体,B cell activating factor receptor, BAFF-R)缺陷型PCP小鼠的肺组织中长链非编码RNA(long non-coding RNA, lncRNA)及信使RNA(messenger RNA, mRNA)的表达谱。此外,为明确差异表达RNA的潜在生物学功能,本研究开展了包含基因本体论(Gene Ontology, GO)富集分析、京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes, KEGG)通路分析以及lncRNA-mRNA共表达网络构建在内的生物信息学分析。本研究进一步探讨了成熟B细胞在PCP发病机制与疾病进展中的作用,结果显示BAFF-R缺陷在PCP模型小鼠群体的免疫调控中发挥重要作用。
