To investigate how HCMV alters monocyte gene expression over time, we collected RNA from HCMV-infected primary human monocytes (3 donors) from 4 hours to 6 weeks post infection and analyzed the differential gene expression by RNAseq. Overall design: Primary human monocytes were mock-infected or HCMV-infected. RNA was collected at 4 hpi, 24 hpi, 2 wpi, 4 wpi, and 6 wpi. 3 replicates are included.
为探究人类巨细胞病毒(Human Cytomegalovirus, HCMV)随时间对单核细胞基因表达的调控模式,我们从3名供体获取原代人单核细胞,对其进行人类巨细胞病毒(HCMV)感染,并在感染后4小时至6周的多个时间点采集总RNA,通过RNA测序(RNAseq)技术分析差异基因表达情况。实验整体设计:将原代人单核细胞分为模拟感染组与HCMV感染组,分别在感染后4小时(4 hpi)、24小时(24 hpi)、2周(2 wpi)、4周(4 wpi)及6周(6 wpi)五个时间点采集RNA样本,每组设置3次生物学重复。
