首先通过电转实验探究MacroH2A敲降后的神经干细胞增殖与分化的情况。发现MacroH2A敲降后促进神经干细胞的增殖,抑制分化,用过体内体外进行Western blot以及免疫荧光染色实验验证这一现象。接着通过转录组测序分析发现MacroH2A的敲低显著下调下游分子NKX2.2。使用CHIP和Co-IP实验证明MacroH2A的敲低下调NKX2.2的表达量,通过乙酰化修饰的方式影响神经干细胞的转录从而影响神经发生的过程。

First, we investigated the proliferation and differentiation of neural stem cells (NSCs) following MacroH2A knockdown via electroporation assays. We observed that MacroH2A knockdown promoted NSC proliferation while inhibiting their differentiation, which was validated by Western blot and immunofluorescence staining experiments both in vitro and in vivo. Subsequently, transcriptome sequencing analysis revealed that MacroH2A knockdown significantly downregulated the expression of the downstream molecule NKX2.2. Chromatin immunoprecipitation (ChIP) and co-immunoprecipitation (Co-IP) assays demonstrated that MacroH2A knockdown downregulated NKX2.2 expression, and affected the transcription of NSCs via acetylation modification, thereby regulating the neurogenesis process.
