Here we have used SSH (Suppression Subtractive Hybridization) approach and generated heat stress responsive ESTs in Indian bread wheat cv. C306 by giving heat stress at two different temperatures. pGEM-T vector was used for cloning and sequenced using T7 primer. The generated data was vecsreen to remove vector and adaptor related contamination.
本研究采用抑制性消减杂交(Suppression Subtractive Hybridization, SSH)技术,以印度普通小麦品种C306为实验材料,通过施加两种不同温度的热胁迫处理,获得了热胁迫响应的表达序列标签(Expressed Sequence Tags, ESTs)。克隆过程使用pGEM-T载体,测序工作通过T7引物完成。所得原始数据经载体筛查(vecscreen)处理,以去除载体及接头相关的污染序列。
