H6H Tropane alkaloid

We downloaded all orthologous sequences of TAa and TPSs of D. stramonium, D. metel and 11 species of Solanaceae (see Table XXXX) to construct the orthogrpups. Downloades sequenceso of proteins and ORF were extracted using TransDecoder and corroborated with GenScan. TPS genes were translated with GenScan, are in the right direction and are the correct CDS; we corroborated this with the NCBI. The database used for BLAST was Uniprot; all available data for the analyzed plant species were downloaded. These amounts 847,544 3. BLAST was carried out with a filtering evalue of 1e-3, and only hits above 85 % were selected.

我们下载了曼陀罗（Datura stramonium）、洋金花（Datura metel）以及11种茄科植物的TAa及萜烯合酶（Terpene Synthases, TPSs）的全部同源序列（详见表XXXX），以构建直系同源基因家族（orthogroups）。使用TransDecoder工具提取所下载的蛋白质序列及开放阅读框（Open Reading Frame, ORF），并通过GenScan基因预测工具进行验证。经GenScan翻译得到的TPS基因序列方向正确且为准确的编码序列（Coding Sequence, CDS），我们已通过美国国家生物技术信息中心（National Center for Biotechnology Information, NCBI）对此进行了核验。本次BLAST（Basic Local Alignment Search Tool）分析所用的数据库为UniProt数据库，我们下载了所分析植物物种的全部可用数据，该数据集共计847,544条序列。BLAST分析设置的过滤E值阈值为1e-3，仅保留比对相似度高于85%的命中结果。
