To investigate the role of Keap1 C151 for induction of gene expression in mouse liver, comprehensive gene expression profiles were compared in the livers of wild-type, Nrf2 knockout, and mice harboring a point mutation at Keap1 C151 (C151S) treated with either CDDO-Me or vehicle Gene expression profiling analysis of RNA-seq data in the livers of three genotypes of mice (wild-type, Nrf2-knockout, Keap1 C151S knock-in) treated with one dose (30 umol/kg BW) of CDDO-Me or vehicle after 24 hours
为探究Kelch样ECH相关蛋白1(Keap1)C151位点在小鼠肝脏基因表达诱导中的调控作用,本研究对经CDDO-Me或赋形剂处理的野生型、核因子红细胞2相关因子2(Nrf2)敲除型以及携带Keap1 C151点突变(C151S)的小鼠的肝脏全面基因表达谱进行了对比分析。同时,本研究对三种基因型小鼠(野生型、Nrf2敲除型、Keap1 C151S敲入型)的肝脏RNA-seq数据开展基因表达谱分析,这些小鼠在单次给予30 μmol/kg体重剂量的CDDO-Me或赋形剂后24小时完成取材。
