To investigate the mechanism how hepatitis B virus (HBV) genome integration into hepatocyte contribute HCC development, we developed HBV integration model where partial HBV genome including HBx region were inserted into TERT promoter region of hiPScells by gene editting. We differenciated the HBV integration model iPS cells into hepatocyte and RNA sequence analysis were performed.

为阐明乙型肝炎病毒(hepatitis B virus, HBV)基因组整合进入肝细胞如何促进肝细胞癌(hepatocellular carcinoma, HCC)发生发展的分子机制,我们构建了HBV整合模型:通过基因编辑技术将包含HBx基因区域的部分HBV基因组插入到人诱导多能干细胞(human induced pluripotent stem cells, hiPSCs)的端粒酶逆转录酶(telomerase reverse transcriptase, TERT)启动子区域内。随后,我们将该HBV整合型诱导多能干细胞诱导分化为肝细胞,并完成了RNA测序分析。
