我们在取自野生型(WT)或Ago4敲除(Ago4 KO)同窝对照小鼠的、经甲型流感病毒(Influenza A)或甲型流感delNS1病毒感染的巨噬细胞中,鉴定出了与AGO(Argonaute蛋白)结合的病毒来源RNA。实验设计:经紫外交联处理后,使用泛AGO抗体对野生型(WT)或Ago4敲除原代骨髓来源巨噬细胞中的AGO结合RNA进行免疫沉淀;上述巨噬细胞分别感染了甲型流感病毒PR/8/1934株或A/PR/8/1934 ΔNS1株,感染复数(MOI)为1,感染时长为24小时。
