Using the highly sensitive RNA array, we assessed the effect of 11β-HSD1 and BVT.2733 on modulating RNAs expression in the neonatal rat ventricular cardiomyocytes. In this study, samples from neonatal rat ventricular cardiomyocytes transfected either with the expression vector for 11β-HSD1 or co-treated with overexpression 11β-HSD1 vector and 11β-HSD1 inhibitor BVT.2733, or transfected with corresponding empty vector were used to acquire the RNA expression profiling thus to elucidate the possible molecular mechanism underlying the effect of 11β-HSD1 on hypertrophy.
本研究采用高灵敏度RNA芯片(RNA array)技术,评估了11β-羟类固醇脱氢酶1(11β-HSD1)与BVT.2733对新生大鼠心室心肌细胞内RNA表达的调控效应。本研究中,我们分别以转染11β-HSD1过表达载体、经11β-HSD1过表达载体与11β-HSD1抑制剂BVT.2733联合处理,以及转染对应空载体的新生大鼠心室心肌细胞样本为实验材料,获取各组的RNA表达谱,以此阐明11β-HSD1调控心肌肥厚的潜在分子机制。
