We report that ITLN1 production is induced by ER stress in IBD, binds to A. muciniphila, regulates mucus thickness, promoting barrier breach and inflammation. RNAseq analysis indicated transcriptional regulation of Itln1 by Xbp1. Overall design: Comparison of gene expression in Paneth cells of XBP1-deleted (Xbp1?IEC) inestinal epithelia of germ-free C57BL/6 mice versus Paneth cells in intestinal epithelia of wild-type germ-free controls.
本研究报道,炎症性肠病(IBD)中的内质网应激(ER stress)可诱导肠凝集素1(ITLN1)的表达;该蛋白可结合嗜黏蛋白阿克曼氏菌(A. muciniphila),调控黏液层厚度,进而促进肠道屏障破坏与炎症反应。RNA测序(RNAseq)分析显示,X盒结合蛋白1(Xbp1)可对Itln1基因实施转录调控。实验设计:对比无菌C57BL/6小鼠XBP1缺失(Xbp1?IEC)肠道上皮中的潘氏细胞(Paneth cells),与野生型无菌对照小鼠肠道上皮中的潘氏细胞的基因表达差异。
