Metastasis is the most important cause of cancer-related death and it has been the most important issue both in basic and clinical cancer researches. Through in vivo spontaneous metastasis animal model, we successfully cultivated and expanded the metastasis lineage cells for genomic, methylomic, and transcriptomic analyses based on the high efficiency and excellent viability of a modified CTC-capturing system together with primary and metastasis tumors as well as parental 4T1-hHER2 breast cancer cell line. Overall design: By using in vivo spontaneous metastasis animal model from NSG mice to harvest all metastatic lineages of cancer cells from primary tumor, circulating tumor cell, and metastasis tumor for following evaluation on cancer metastasis evolution.
肿瘤转移是癌症相关死亡的最主要诱因,同时也是基础与临床癌症研究领域的核心议题。本研究借助经改造的循环肿瘤细胞(Circulating Tumor Cell, CTC)捕获系统所具备的高效性与优异活性,结合原发肿瘤、转移瘤以及亲本4T1-hHER2乳腺癌细胞株,通过体内自发转移动物模型成功培养并扩增了转移谱系细胞,用于基因组、甲基化组及转录组相关分析。实验整体设计:利用NSG小鼠构建体内自发转移动物模型,从原发肿瘤、循环肿瘤细胞及转移瘤中分离获取全部癌症细胞转移谱系,用于后续肿瘤转移演进过程的相关评估。
