Genomewide DNA methylation array profiling of 23 previously unpublished CNS tumors resolved into a new entity, high-grade glioma with pleomorphic and pseudopapillary features (HPAP). Tumors demonstrate a variety of histologic diagnoses including glioblastoma (GBM), pleomorphic xanthoastrocytoma (PXA), anaplastic ependymoma, and polymorphous low-grade neuroepithelial tumor of the young (PLNTY). The Illumina Infinium EPIC 850k Human DNA Methylation Beadchip was used to obtain DNA methylation profiles across approximately 850,000 CpG sites of genomic DNA extracted from formalin-fixed, paraffin-embedded tumor tissue. 本数据集针对23例此前未被报道的中枢神经系统(Central Nervous System, CNS)肿瘤开展全基因组DNA甲基化芯片分析,相关肿瘤被归为一种新型实体亚型:伴多形性与假乳头状特征的高级别胶质瘤(high-grade glioma with pleomorphic and pseudopapillary features, HPAP)。 该队列肿瘤涵盖多种组织学诊断类型,包括胶质母细胞瘤(glioblastoma, GBM)、多形性黄色星形细胞瘤(pleomorphic xanthoastrocytoma, PXA)、间变性室管膜瘤以及青年多形性低级别神经上皮肿瘤(polymorphous low-grade neuroepithelial tumor of the young, PLNTY)。 本研究采用Illumina Infinium EPIC 850K人类DNA甲基化芯片(Illumina Infinium EPIC 850k Human DNA Methylation Beadchip),对从福尔马林固定石蜡包埋肿瘤组织中提取的基因组DNA进行检测,获取约85万个CpG位点的DNA甲基化谱。 实验整体设计:将23份肿瘤样本提取的亚硫酸氢盐转化基因组DNA,与Illumina Infinium EPIC人类DNA甲基化芯片进行杂交。
