TO explore role of DGCR8, a essential factor in canonical microRNAs biogenesis, in intestinal epithelial regeneration, we performed gene expression profiling analysis using data obtained from RNA-seq of intestinal organoids, intesitnal stem cells, and crypts from mice following 5-FU administration. To further investigate which microRNA was responsible for intestinal regeneration, we also performed small RNA sequencing with intestinal stem cells and intestinal epithelia cells. Overall design: Comparative transcriptome profiling analysis of RNA-seq data for intestinal stem cells, intestinal organoids, and crypts from mice following 5-FU administration.
为探究经典微RNA(microRNAs)生物发生过程中的关键因子DGCR8在小肠上皮再生中的作用,我们利用经5-氟尿嘧啶(5-FU)处理的小鼠小肠类器官、小肠干细胞及隐窝的RNA测序(RNA-seq)数据开展了基因表达谱分析。为进一步明确参与小肠再生调控的微RNA种类,我们还针对小肠干细胞与小肠上皮细胞进行了小RNA测序(small RNA sequencing)。整体实验设计:对经5-FU处理的小鼠小肠干细胞、小肠类器官及隐窝的RNA测序数据进行比较转录组分析。
