Cancer cachexia is a metabolic syndrome with alterations in gene expression profile that consequently lead to skeletal muscle wasting. Here, we used RNA sequencing to analyze the mRNA expression profile in the tibialis anterior muscles of the Lewis lung carcinoma (LLC) model of cancer cachexia. Overall design: Eight-week-old healthy C57BL/10 male mice were inoculated subcutaneously with a total of 1.5 x106 LLC cells (7.5 x 105 cells in 0.1 mL PBS, in each flank). Ten mice (control group) were injected with equal volumes of 1X PBS. LCC and control mice were studied after 22 days after LLC cells inoculation when the LLC group had developed overt cachexia.
癌症恶病质(Cancer cachexia)是一类伴随基因表达谱改变的代谢综合征,可最终引发骨骼肌消耗。本研究采用RNA测序(RNA sequencing)技术,对路易斯肺癌(Lewis lung carcinoma, LLC)癌症恶病质模型小鼠的胫骨前肌组织mRNA表达谱进行分析。实验设计:选取8周龄健康C57BL/10雄性小鼠,于双侧侧腹皮下接种总计1.5×10⁶个LLC细胞(每侧接种0.1mL含7.5×10⁵个细胞的1×磷酸盐缓冲液(PBS));对照组10只小鼠注射等体积的1×PBS。在LLC组小鼠出现明显恶病质时,于接种LLC细胞后22天对两组小鼠开展分析研究。
